We have characterized the rat 5-LO gene promoter region, and investigated the regulatory mechanisms involved in the expression of 5-LO in macrophages and epithelial cells. Transcriptional start sites were determined and 5-LO promoter fragment (-1676 to +72) was cloned into the pGL3-basic vector. Transient transfection experiments indicated that the nucleotides -1145 to -1136, -1066 to -1056 and -1025 to -1006 were essential for basal transcription of the 5-LO gene in both macrophages and epithelial cells. Binding of nuclear proteins to the region between -1006 and -771 and the region between -771 and -376 is essential for basal transcription. Competitive assays indicated that both protein(s) binding to the region between -771 and -376 and factor(s) binding to the region between -1025 and -1006 are macrophage-specific.
